In thin-layer chromatography, the separation of the components of a mixture is the most delicate step, as it is subject to different and variable parameters.
Some are perfectly controlled (chemical nature of the stationary and mobile phase), others are more random (degree of humidity, ambient temperature, pre-saturation).
All require precise and rigorous protocols to achieve correct, repeatable and reproducible separations. The quality of the migration separation chamber and the choice of its size and shape are very important.

Standard TLC Developing Chamber biostep® (DESAGA)
Special features of this separating chamber are the flat edge of the flange and the grooved base of the chamber. The glass button cover seals the separating chamber.
After the sample and/or standard has been applied, the separation process takes place in order to separate the components of the applied substances. To achieve this goal, the separation chambers must fulfil several requirements:
- They must contain the mobile phase and bring it to the absorbent layer.
- They must be fitted with a tight-fitting lid to ensure that the gas chamber can be saturated with solvent vapours if necessary (working with chamber saturation).
- The front of the solvent must be observed, which is why a transparent material is important.
- The material should be chemically resistant and easy to clean.
The standard TLC separation chamber fulfils all these requirements. It is usually used in conjunction with a TLC filter paper to achieve better and faster saturation of the tank’s atmosphere.

Although the principle of thin-layer chromatography is more than a century old, it did not make its breakthrough as an analytical method until about 50 years ago.
Thanks to the development of new sorbents and supports, as well as increasing instrumentation and automation, TLC has become a versatile separation method. It is used both in qualitative analysis and in quantitative analysis.
Applications range from simple manual separation processes in classic TLC to automated processes in HPTLC (high performance thin layer chromatography).
Advantages of thin layer chromatography:
