Tip:
A well-known method for protein sequencing is the Edman degradation. Herein, the peptide is degraded using phenylisothiocyanate (Art. No. 7296) starting from the N-terminus. Phenylisothiocyanate reacts with the N-terminal amino acid resulting in cleavage from the peptide and formation of a phenylthiohydantoin derivative. Subsequently, the phenylthiohydantoin derivative is identified by chromatographic methods through comparison with standards and the next amino acid can be addressed. Through repetition of this procedure stepwise degradation of the peptide is achieved, thus enabling sequencing of peptides with up to 50 residues.
