N,N-Dimethylformamide (DMF) is a colourless, polar, aprotic solvent with a high boiling point that is frequently used in peptide synthesis. It is known for its ability to dissolve a wide range of organic and inorganic compounds.


In peptide synthesis, DMF is used to dissolve amino acids and peptide coupling reagents such as N,N'-dicyclohexylcarbodiimide (DCC), which is essential for the formation of peptide bonds. DMF is also commonly used as a solvent for other reagents and additives used in peptide synthesis, e.g. bases such as diisopropylethylamine (DIPEA). The polar, aprotic nature of DMF makes it an excellent solvent for reactions involving polar or charged species. It can also act as a dehydrating agent, helping to remove water from reaction mixtures, which is essential in peptide synthesis where water can compete with the formation of peptide bonds.
