RNA-ClickSeq Library Prep Kit
ClickSeq is a simple method for synthesizing NGS libraries using click-chemistry instead of traditional ligation enzymes to attach sequencing adapters—a step often prone to errors. During reverse transcription, 3′-azido-nucleotides are incorporated, which terminate the growing cDNA strands. These azido-blocked cDNAs are then click-ligated to alkyne-modified adapters and PCR-amplified to generate sequencing-ready libraries.ClickSeq is a flexible and modular platform that supports both RNA and DNA inputs. By avoiding enzymatic ligation and fragmentation, the protocol is faster and more efficient than standard RNA-seq workflows.

General information
The copper(I)-catalysed azide-alkyne cycloaddition (CuAAC) is the most prominent example of a group of reactions named click reactions. According to Sharpless’ definition, these reactions are characterised by high yields, mild reaction conditions, and by their tolerance of a broad range of functional groups.[1-3] Typically, the reactions require simple or no workup or purification of the product. The most important characteristic of the CuAAC reaction is its unique bioorthogonality as neither azide nor terminal alkyne functional groups are generally present in natural systems.
